Until now, researchers interested
in very rare transcripts have been unable to take advantage of standard
cDNA-based methodologies. In conventional cDNA synthesis methods,
high- to medium-abundance transcripts are reverse-transcribed with
relative ease; however, rare transcripts are not easily converted
to full-length cDNA. We designed the proprietary Gene Enrichment Technology
(GET; patent pending) to specifically enrich for full-length, rare
transcripts in the final cDNA preparation. Thus, researchers can rapidly
and easily amplify almost any target transcript-regardless of its
abundance.
Each GETRare Full-Length cDNA is tissue-specific, double-stranded
cDNA that has already been pre-enriched for rare transcripts (Figure
1). You can immediately get results with this ready-to-use cDNA-just
run a simple PCR and visualize your results by agarose gel electrophoresis.
GETRare Full-Length cDNAs are ideal
for amplifying your gene of interest using gene-specific
primers. Alternatively, you can profile expression of your gene of
interest by using GETRare Full-Length cDNAs prepared from different
tissues.
Figure. Rare, full-length genes were successfully amplified using
Human Testis GETRare Full-Length cDNA compared to that of other commercially
available, non-enriched cDNA. Lanes 1-3: Human Testis Full-Length
cDNA. Lanes 4-6: Competitor, non-enriched cDNA. Lanes 1 & 4: IL-10.
Lanes 2 & 5: p50. Lanes 3 & 6: IGFR. Lane M: 1-kb DNA ladder.
